电压门控钙离子通道(voltage-gated calcium channels, VGCCs)构象变化主要取决于膜电位的变化从而使通道呈现开放或关闭状态,控制钙离子的流动。电压门控钙离子通道作为可兴奋性细胞膜表面的主要钙离子通道,对胞内钙信号的强弱和动力学有重要的调节作用,其主要功能是将细胞电活动转化为钙离子信号继而激活下游信号通路,发挥重要的生理功能,如神经系统的发育、神经递质释放和突触可塑性等。因此,研究电压门控钙离子通道的调控特性对认识生理活动中胞内钙信号具有重要意义。
智力发育障碍的分子机制:Rett综合征是严重影响儿童精神运动的一种神经发育性遗传病。鉴于MeCP2功能与BDNF的mRNA和蛋白表达水平之间复杂的关系,从BDNF的转录调控、分泌、可释放BDNF量和胞外调控等多个层次上研究Rett综合征动物模型(MeCP2基因敲除小鼠)其海马神经元内源性BDNF释放的作用及其机制,有望阐明内源性BDNF的释放是否是Rett综合征发病的病理生理学机制,揭示智力障碍和孤独症相关疾病的基本机制并为其提供可能的治疗策略。
